Mike Rounds was born in Huron, South Dakota, on October 24, 1954, to Joyce and Don Rounds. He is descended from Germans, Belgians, Swedes, and English. Since he was three years old, Rounds has lived in Pierre, the state capital. Marion Rounds, his uncle, was killed in the Pacific theatre of World War II and was named after him. Several members of the Rounds family have served in the Texas legislature.

His father served as state director of highway safety, a Rural Electrification Administration worker, and executive director of the South Dakota Petroleum Council, among other positions. Tim Rounds, his brother, is a member of the South Dakota Legislature, representing Pierre’s District 24. Rounds obtained his bachelor’s degree in political science from South Dakota State University in Brookings.

Rounds met his wife, Jean, from Lake Preston, South Dakota, while attending South Dakota State University. They have four children and were married in 1978. Tim Rounds’ older brother is him.

Rounds was a former partner of Fischer Rounds & Associates, a real estate and insurance firm with offices in Pierre, Rapid City, Mitchell, Watertown, and Sioux Falls.

As the contest for governor in 2002 began to take form, the media and political pundits widely regarded Rounds as a long shot. John Thune, then-Congressman, was the front-runner for the nomination until late 2001. When Thune dropped out of the fight to unseat Senator Tim Johnson, state Attorney General Mark Barnett and former Lieutenant Governor Steve T. Kirby stepped in to fill the void.

Rounds was rumoured to be running for the United States Senate seat now held by Tim Johnson, a Democrat who had served since 1997. Johnson decided not to seek re-election.

Rounds announced his candidacy for the seat vacated by Johnson’s retirement on November 29, 2012. He defeated four other candidates in the Republican primary in June 2014. Early surveys showed Rounds leading Democratic opponent Rick Weiland by a 2–1 margin. Rounds, Weiland, and independent former Senator Larry Pressler were in a tighter three-way race in October 2014 polls. Gordon Howie, a former state legislator who is an independent conservative, was also a candidate.

Rounds was one of 20 senators who sponsored the Employer Participation in Repayment Act in February 2019, which allows employers to contribute up to $5,250 to their employees’ student loans as a way of providing relief and encouraging people to apply for jobs with companies that implement the policy.

Rounds won the Republican nomination in 2020 after defeating Scyller Borglum. He defeated Democrat Dan Ahlers in the general election.
