Amy Louise Acker was born in Dallas in 1976, Texas to a stay-at-home mom and a lawyer dad. For fourteen years, she pursued training in ballet and modern dance. Her ballet career was cut short when she had knee surgery as a high school student.  Acker is a Lake Highlands High School alumnus from the DFW area.

The next year, she graduated with honors from Southern Methodist University with a bachelor’s degree in drama. Acker walked the runway for J. Crew during her sophomore year of college. In the same year, she also earned a Bachelor of Fine Arts degree.

For several seasons, she performed as a theatre performer, including a stay at the American Players Theatre in Spring Green, Wisconsin. Acker married actor James Carpinello in Los Angeles on April 25, 2003. In February 2005, they gave birth to a son, and in September 2006, they had a daughter.

Acker made her big-screen debut as Winifred “Fred” Burkle in Angel (seasons 2–5), and she also played Illyria for part of the fifth and final season of the show. For her role, she was nominated for a Saturn Award in 2003 for Best Supporting Actress in a TV Movie or Miniseries. In Alias’ last season in 2005, Acker joined the cast as Kelly Peyton, the show’s villain.

As the series’ last episodes aired in April and May of 2006, Acker made the transition from guest star to a regular cast member. Acker also lent her voice to the animated series Justice League Unlimited as Huntress in 2005.

Later, she appeared as a special guest on How I Met Your Mother in which she reconnected with Angel co-star Alexi Denisof (the real-life husband of HIMYM actor Alyson Hannigan).

In an episode of Marvel’s Agents of S.H.I.E.L.D., Acker played Audrey, Phil Coulson’s former flame. Acker played Samantha “Root” Groves on the CBS drama Person of Interest from 2012 to 2016. Groves was introduced as a regular character in the third season of the show. Acker became the Machine’s voice actor starting with the 100th episode of the show.

She also appears as the Machine’s visual manifestation in the series finale. Acker was cast as Caitlin Strucker in the Fox pilot The Gifted in March 2017, and the show was renewed for a second season in May of the same year. Fox canceled The Gifted after two seasons on April 17, 2019, two months after it premiered on October 2, 2017.

In 2019, Acker co-starred with Fred Savage in the aftershow parody What Just Happened??!. In the “show-within-a-show,” a fictional drama series called The Flare, she plays Dr. Rachel Layne.
